    Abstract [en]

    The development of long-lasting zirconia-based ceramics for implants, which are not prone to hydrothermal aging, is not satisfactorily solved. Therefore, this study is conceived as an overall evaluation screening of novel ceria-stabilized zirconia-alumina-aluminate composite ceramics (ZA8Sr8-Ce11) with different surface topographies for use in clinical applications. Ceria-stabilized zirconia is chosen as the matrix for the composite material, due to its lower susceptibility to aging than yttria-stabilized zirconia (3Y-TZP). This assessment is carried out on three preclinical investigation levels, indicating an overall biocompatibility of ceria-stabilized zirconia-based ceramics, both in vitro and in vivo. Long-term attachment and mineralized extracellular matrix (ECM) deposition of primary osteoblasts are the most distinct on porous ZA8Sr8-Ce11p surfaces, while ECM attachment on 3Y-TZP and ZA8Sr8-Ce11 with compact surface texture is poor. In this regard, the animal study confirms the porous ZA8Sr8-Ce11p to be the most favorable material, showing the highest bone-to-implant contact values and implant stability post implantation in comparison with control groups. Moreover, the microbiological evaluation reveals no favoritism of biofilm formation on the porous ZA8Sr8-Ce11p when compared to a smooth control surface. Hence, together with the in vitro in vivo assessment analogy, the promising clinical potential of this novel ZA8Sr8-Ce11 as an implant material is demonstrated.

    Abstract [en]

    Objective To adequately address clinically important issues such as osseointegration and soft tissue integration, we screened for the direct biological cell response by culturing human osteoblasts and gingival fibroblasts on novel zirconia-based dental implant biomaterials and subjecting them to transcriptional analysis. Methods Biomaterials used for osteoblasts involved micro-roughened surfaces made of a new type of ceria-stabilized zirconia composite with two different topographies, zirconium dioxide, and yttria-stabilized zirconia (control). For fibroblasts smooth ceria- and yttria-stabilized zirconia surface were used. The expression of 90 issue-relevant genes was determined on mRNA transcription level by real-time PCR Array technology after growth periods of 1 and 7 days. Results Generally, modulation of gene transcription exhibited a dual dependence, first by time and second by the biomaterial, whereas biomaterial-triggered changes were predominantly caused by the biomaterials’ chemistry rather than surface topography. Per se, modulated genes assigned to regenerative tissue processes such as fracture healing and wound healing and in detail included colony stimulating factors (CSF2 and CSF3), growth factors, which regulate bone matrix properties (e.g. BMP3 and TGFB1), osteogenic BMPs (BMP2/4/6/7) and transcription factors (RUNX2 and SP7), matrix collagens and osteocalcin, laminins as well as integrin ß1 and MMP-2. Significance With respect to the biomaterials under study, the screening showed that a new zirconia-based composite stabilized with ceria may be promising to provide clinically desired periodontal tissue integration. Moreover, by detecting biomarkers modulated in a time- and/or biomaterial-dependent manner, we identified candidate genes for the targeted analysis of cell-implant bioresponse during biomaterial research and development.

    Abstract [en]

    Hydroxypropylated and oxidised potato starch (HONPS) was used together with glycerol and water to produce thermoplastic starch. The amount of glycerol was kept constant at 22 parts by weight per 100 parts of dry starch. The thermoplastic starch was converted into films/sheets using three different processing techniques; casting, compression moulding and film blowing. The last two methods represent typical thermoplastic conversion techniques requiring elevated processing temperatures. By means of size-exclusion chromatography, it was found that compression moulding and film blowing led to some degradation of high-molecular weight amylopectin as well as of high-molecular weight amylose-like molecules. The degradation was significantly less pronounced for the cast films. The morphology of the specimens was quite complex and phase separations on different levels were identified. In the cast films and, to a lesser extent, in the compression-moulded specimens, a fine network structure could be distinguished. Such a structure could however not be ascertained in the film-blown material and this is discussed in terms of the thermo-mechanical treatment of the starch materials. © 2007 Elsevier Ltd.     Abstract [en]

    We study the problem of specifying and verifying properties of pi-calculus processes while relying on a bisimulation semantics. As our property specification language we use a version of the modal mu-calculus adapted to the pi-calculus. We show that the logical language is sufficiently expressive to characterize by means of a finite formula a process up to any approximation of the bisimulation relation. We consider the problem of checking that a process of the pi-calculus satisfies a specification expressed in this modal mu-calculus. We develop an algorithm which is sound in general, and complete for processes having a finite reachability property. Finally, we present a proof system which can be applied to prove non-recursive properties of arbitrary processes. We show that the system is complete on the non-recursive fragment of the logical language.

    Abstract [en]

    We address the specification and verification problem for process calculi such as Chocs, CML and Facile where processes or functions are transmissible values. Our work takes place in the context of a static treatment of restriction and of a bisimulation-based semantics. As a paradigmatic and simple case we concentrate on (Plain) Chocs. We show that Chocs bisimulation can be characterized by an extension of Hennessy-Milner logic including a constructive implication, or function space constructor. This result is a non-trivial extension of the classical characterization result for labelled transition systems. In the second part of the paper we address the problem of developing a proof system for the verification of process specifications. Building on previous work for CCS we present an infinitary sound and complete proof system for the fragment of the calculus not handling restriction.

    Generally, there is a trade-off between methods of gene expression analysis that are precise but labor-intensive, e.g. RT-PCR, and methods that scale up to global coverage but are not quite as quantitative, e.g. microarrays. In the present paper, we show how how a known method of gene expression profiling (K. Kato, Nucleic Acids Research 23, 3685-3690 (1995)), which relies on a fairly small number of steps, can be turned into a global gene expression measurement by advanced data post-processing, with potentially little loss of accuracy. Post-processing here entails solving an ancillary combinatorial optimization problem. Validation is performed on in silico experiments generated from the FANTOM data base of full-length mouse cDNA. We present two variants of the method. One uses state-of-the-art commercial software for solving problems of this kind, the other a code developed by us specifically for this purpose, released in the public domain under GPL license.

    Abstract [en]

    We propose a new facile electrochemical sensing platform for determination of urea, based on a glassy carbon electrode (GCE) modified with nickel cobalt oxide (NiCo 2 O 4 ) nanoneedles. These nanoneedles are used for the first time for highly sensitive determination of urea with the lowest detection limit (1 μM) ever reported for the non-enzymatic approach. The nanoneedles were grown through a simple and low-temperature aqueous chemical method. We characterized the structural and morphological properties of the NiCo 2 O 4 nanoneedles by TEM, SEM, XPS and XRD. The bimetallic nickel cobalt oxide exhibits nanoneedle morphology, which results from the self-assembly of nanoparticles. The NiCo 2 O 4 nanoneedles are exclusively composed of Ni, Co, and O and exhibit a cubic crystalline phase. Cyclic voltammetry was used to study the enhanced electrochemical properties of a NiCo 2 O 4 nanoneedle-modified GCE by overcoming the typical poor conductivity of bare NiO and Co 3 O 4 . The GCE-modified electrode is highly sensitive towards urea, with a linear response (R 2 = 0.99) over the concentration range 0.01-5 mM and with a detection limit of 1.0 μM. The proposed non-enzymatic urea sensor is highly selective even in the presence of common interferents such as glucose, uric acid, and ascorbic acid. This new urea sensor has good viability for urea analysis in urine samples and can represent a significant advancement in the field, owing to the simple and cost-effective fabrication of electrodes, which can be used as a promising analytical tool for urea estimation.

    A Virtual Design Studio for Low-Frequency Sound from Walking in Lightweight Buildings2019Doctoral thesis, monograph (Other academic)
    Abstract [en]

    In recent years there has been a growing interest for building lightweight multistorey woodenresidential buildings in countries like Sweden with large and renewable forests. While positive aspectsof these buildings, such as sustainability, ease of construction and lightness, motivate building more inwood, poor acoustic performance is a risk which concerns the wooden-building industry.Low-frequency impact sound from walking of the neighbors upstairs is the main source of complaintsabout the acoustic performance of these buildings. The disturbance caused by walking sounds,transmitted through lightweight wooden floors, results in acoustic discomfort and impairs theperceived quality of the building; sometimes even when the building has fulfilled an acoustic classhigher than minimum requirement, according to the national standard on sound classification and itssingle number ratings. The standard methods for objective evaluation of impact sound insulation offloors cannot predict, at a satisfactory level, the walking sound annoyance that the inhabitants ofwooden buildings experience. This causes an uncertainty about the resulting perceived quality of thesebuildings, which greatly concerns the building manufacturers and demotivates them from choosinglightweight wooden elements over heavyweight building materials such as concrete. This uncertaintycan be overcome by evaluating the perceived acoustic quality of the building prior to its construction.

    One solution is to build test houses where the subjective acoustic performance of floor samples can beevaluated in advance to the building construction. However, building a test house is expensive;besides, for evaluating the effect of every design modification on the experienced acoustic comfort ofthe building, a real floor sample has to be built and installed in the house, which would be timeconsumingand costly. An alternative solution is to use virtual acoustic test facilities.In this thesis a virtual design studio for impact sound is developed. It is a tool that facilitates creatingand listening to the acoustic field generated by impact forces such as footsteps on lightweight floors. Italso provides the possibility to evaluate the acoustic performance of floor elements in an early designphase, and to investigate the correlation between design parameters and the perceived impact soundinsulation of the floor. The tool is demonstrated and a very first listening test shows that one can obtainresults which are in good agreement with the results in literature. Loudness, reverberation andthumping are shown to influence the annoyance. It is also shown that there is a difference in judgementof walking sounds by persons who have experience with lightweight floors at home and by those whodo not have that experience.

    Measurement and modeling of low frequency ground reaction forces (GRFs) from human walking have been the subjects of research in different fields from biomechanics to civil engineering and structural dynamics. Many of the existing models are developed based on experiments which alter natural walking by for example presence of force transducers, limitations in the speed and path of walking and replacing the real floor with a transducerfacilitated measurement rig. These alterations result in contact forces which do not represent real GRFs. In this study, a time-domain inverse measurement method based on LMS algorithm is used to measure low-frequency (<100 Hz) forces induced by human footsteps. The LMS-based force identification method is first validated for low-frequency excitations with less complexity in number of excitation positions and frequency content compared with the footsteps. The method is then applied to measure ground reaction forces created by human walking.

    Despite many advantages of wooden buildings, low-frequency noise disturbance, mainly from human footsteps on timber floors, is a major hindrance for widespread application of wood in multi-storey buildings. In many cases the impact noise evaluations of the wooden floors according to ISO 140-7 and ISO 717-2 standards do not correspond to subjective experiences. This study aims to investigate the characteristics of footstep forces to support further studies related to discrepancies between standard evaluations and low-frequency noise disturbances in lightweight buildings. Transient footstep forces in vertical direction made by different walkers and different footwear on two floor structures were measured and compared. The vertical footstep forces were then compared with the impact forces made by a standard tapping machine. Analysis revealed that footstep forces contain large amplitudes at low frequencies (20 < f < 50) which are not included in the standard evaluation procedure. Therefore, by extending the frequency range of the standard measurements down to 20 Hz and designing an accurate adaptation term for evaluations, a better correlation between standard impact noise evaluations and acoustic performance of the lightweight buildings might be achieved.

    Abstract [en]

    Interfacial properties of two brush-with-anchor mucins, C-P55 and C-PSLex, have been investigated at the aqueous solution/poly(methyl methacrylate) (PMMA) interface. Both are recombinant mucin-type fusion proteins, produced by fusing the glycosylated mucin part of P-selectin glycoprotein ligand-1 (PSLG-1) to the Fc part of a mouse immunoglobulin in two different cells. They are mainly expressed as dimers upon production. Analysis of the O-glycans shows that the C-PSLex mucin has the longer and more branched side chains, but C-P55 has slightly higher sialic acid content. The adsorption of the mucins to PMMA surfaces was studied by quartz crystal microbalance with dissipation. The sensed mass, including the adsorbed mucin and water trapped in the layer, was found to be similar for these two mucin layers. Atomic force microscopy with colloidal probe was employed to study surface and friction forces between mucin-coated PMMA surfaces. Purely repulsive forces of steric origin were observed between mucin layers on compression, whereas a small adhesion was detected between both mucin layers on decompression. This was attributed to chain entanglement. The friction force between C-PSLex-coated PMMA is lower than that between C-P55-coated PMMA at low loads, but vice versa at high loads. We discuss our results in terms of the differences in the glycosylation composition of these two mucins.

    Equilibration in adsorbing polymer systems can be very slow, leading to different physical properties at a given condition depending on the pathway that was used to reach this state. Here we explore this phenomenon using a diblock copolymer consisting of a cationic anchor block and a thermoresponsive block of poly(2-isopropyl-2-oxazoline), PIPOZ. We find that at a given temperature different polymer chain densities at the silica surface are achieved depending on the previous temperature history. We explore how this affects surface and friction forces between such layers using the atomic force microscope colloidal probe technique. The surface forces are purely repulsive at temperatures <40 °C. A local force minimum at short separation develops at 40 °C and a strong attraction due to capillary condensation of a polymer-rich phase is observed close to the bulk phase separation temperature. The friction forces decrease in the cooling stage due to rehydration of the PIPOZ chain. A consequence of the adsorption hysteresis is that the friction forces measured at 25 °C are significantly lower after exposure to a temperature of 40 °C than prior to heating, which is due to higher polymer chain density on the surface after heating.

    Abstract [en]

    Thermoresponsive polymer layers on silica surfaces have been obtained by utilizing electrostatically driven adsorption of a cationic–nonionic diblock copolymer. The cationic block provides strong anchoring to the surface for the nonionic block of poly(2-isopropyl-2-oxazoline), referred to as PIPOZ. The PIPOZ chain interacts favorably with water at low temperatures, but above 46 °C aqueous solutions of PIPOZ phase separate as water becomes a poor solvent for the polymer. We explore how a change in solvent condition affects interactions between such adsorbed layers and report temperature effects on both normal forces and friction forces. To gain further insight, we utilize self-consistent lattice mean-field theory to follow how changes in temperature affect the polymer segment density distributions and to calculate surface force curves. We find that with worsening of the solvent condition an attraction develops between the adsorbed PIPOZ layers, and this observation is in good agreement with predictions of the mean-field theory. The modeling also demonstrates that the segment density profile and the degree of chain interpenetration under a given load between two PIPOZ-coated surfaces rise significantly with increasing temperature.

    Abstract [en]

    Several attempts have been made in the past to develop a European harmonized testing and assessment method for façades before the European commission decided to publish a call for tender on the topic. A project consortium from five countries (Sweden, UK, France, Germany and Hungary) applied to the call for tender and was contracted to develop a European approach to assess the fire performance of façades. 24 sub-contractors and 14 stakeholder entities were part of the project.

    The objective of the European project was to address a request from the Standing Committee of Construction (SCC) to provide EC Member States regulators with a means to regulate the fire performance of façade systems based on a European approach agreed by SCC. In addressing this objective, the project team was asked to consider a number of issues which are presented and discussed.

    The initial stages of this project were focused on:

    • establishing a register of the regulatory requirements in all Member States in relation to the fire performance of façade systems, and
    • to identify those Member States who have regulatory requirements for the fire performance façade systems which go beyond the current EN 13501 (reaction to fire and fire resistance) classification systems and to collate the details of these additional requirements. [6]

    After having confirmed the regulatory needs the following steps were discussed:

    • a testing and classification methodology based on BS 8414 - Fire performance of external cladding systems series and DIN 4102-20 - Fire behaviour of building materials and building components - Part 20: Complementary verification for the assessment of the fire behaviour of external wall claddings to address the identified key performance and classification characteristics [4] [5]
    • a verification and validation proposal, in the form of a round robin programme to support the development of the proposed testing and classification methodologies.
    • an alternative test method which was developed on the basis of the comments from stakeholders during the project

    Several hundreds of comments were received during the project and were implemented in the development.

    This paper is a short overview of results the two-year development work, which Final Report published by the European Commission in 2018 [1].

    Abstract [en]

    Façade fires do not occur often (in comparison to other major structure fires) but in recent years there have been a number of spectacular façade fires in high rise building such as the recent fire in Grenfell Tower, London.Under-ventilated compartment fires may cause flames to spill out of window openings impinging the façade, thus devastating façade fires may start on one floor leap-frogging to adjacent floors. It is therefore necessary to limit or delay fire spread to higher floors. Requirements built on large scale fire testing may decrease the risk of these types of fires provided that the building is constructed according to the specifications provided by the manufacturer. Different countries have different regulations and tests for façades. New materials and façade systems are continuously introduced which might call for an update of these tests and regulations.This report summarizes experimental and modelling efforts in characterizing the fire safety of façades using the Swedish SP Fire 105 and the British BS 8414 methods. Recent experimental results and modelling is presented exploring the variations in the fire exposure, fire load and the fuel used. The fire source and the heat exposure to the façade are characterized by additional temperatures measured by plate thermometers while some other aspects are only treated in the numerical study such as a change in fuel. It is found that the results from the BS 8414 are largely affected by wind and climate since the experimental test was performed outdoors, moreover fire spread on wooden façades is also briefly discussed.In order to obtain a deeper understanding of the test methods and the results CFD (Computational Fluid Dynamics) Modelling in FDS was used. The models were based on measured input parameters including uncertainties and an assessment of the impact of said uncertainties. The models could often reproduce the experimentally found temperatures qualitatively and quantitatively. A detailed discussion on the regulations and the tests that lead to the SP Fire 105 test method is also presented. Summaries of the façade testing methods and conditions in other European countries are presented in the appendices.Finally possible ways forward in updating the façade testing and regulations are discussed.

    A comparison between full‐scale façade fire tests where SP Fire 105 and BS 8414‐1 were used regarding repeatability and the use of modeling to discern changes in the setups is presented. Two test series according to BS 8414‐1 were repeated outside using the same façade systems on 2 different days, whereas for the SP Fire 105 a set of common façade systems in Sweden were tested indoors. In particular, the results show that the wind around the test setup may have a significant impact on the tests and that the heat exposure to the façade surface will depend on the thickness of the test specimen where an increased temperature in front of the façade, and a decreased temperature on the façade 2.1 m above the fire room, is observed experimentally. The heat exposure to the test specimen varies to a more limited extent when an uncontrollable free burning fire source is used (in this study heptane and wood, respectively) and that this variation increases when wind is present.

    In this paper, a comparative simulation study on 3 large‐scale facade testing methods, namely,the SP Fire 105, BS 8414‐1, and the ISO 13785‐2 methods, is presented. Generally goodcorrespondence between simulations and experimental data has been found, provided thatthermal properties of the facade material and heat release rates are known; however, thecorrespondence deviates in close proximity of the fire source. Furthermore, a statistical ensemblefor evaluating the effects stemming from uncertainty in input data is used. Here, it wasfound using this statistical ensemble that the variability was smaller in the ISO 13785‐2compared to the BS 8414‐1 method. The heat release rates (HRR) used in the simulations wereadopted from measurements except for the ISO method where the information in the standardwas used to approximate the HRR. A quantitative similarity between the HRR in the ISOmethod and the British method was found.

    Abstract [en]

    In this paper a comparison between test and modelling results are performed for two large-scale façade fire testing methods, namely SP Fire 105 and BS 8414-1. In order to be able to compare tests and modelling the uncertainties have to be quantified both in the test and the modelling. Here we present a methodology based on deterministic sampling to quantify uncertainties in the modelling input. We find, in general good agreement between the models and the test results. Moreover, temperatures estimated by plate thermometers is indicated to be less sensitive to small variations in model input and is thus suitable for these kind of comparisons.
